EurekAlert!
We showed that the extremely thermophilic bacterium Thermus thermophilus (isolated from places such as hot springs) moves against water flow. This is called positive rheotaxis. When nutrients run out, the cells stand up with one pole still attached to the solid surface and move upstream in that position. Watch the video here. We found that this response is common among many bacteria closely related to this species. Naoki did a lot of the experiments. Students also received several presentation awards: 1, 2, 3, 4, 5, 6, 7, 8. The work was featured in a UEC news release, and we also posted it on EurekAlert!. It was even selected as an Editor's Choice!

We found that Mycoplasma, a small parasitic bacterium, shows rheotaxis. Rheotaxis is the ability to steer one's direction of movement in response to flow. Fish are known to swim against the current, and it seems that Mycoplasma, the smallest living organism, has a similar ability. Mycoplasma is well known to live as a parasite on higher organisms, including humans, and it probably makes clever use of the fluid flow in these real environments to reach its destination. Kabata, a student in my group when I was at Gakushuin University, did a lot of the experiments. Many thanks. Yoshiki received the Student Presentation Award at the Annual Meeting of the Biophysical Society of Japan. The press release for this paper is here.

We showed that in the thermophilic cyanobacterium Thermosynechococcus, switching between positive and negative phototaxis is controlled by a molecule called c-di-GMP. When exposed to light, the cells create a polarity along their short axis and distribute type IV pili asymmetrically, which produces directional movement. Observing at 45 °C was tricky in places, but the mutants made by Gen in Freiburg were perfect, and we were able to visualize the dynamics of type IV pilus filaments only about 7 nm thick. Working together with Annegret across Germany and Japan, we got a wonderful result. The press release for this paper is here. It was also selected for an eLife Digest . Gen explains the work on Twitter.

We found that a pathogenic bacterium that causes human pneumonia has a protein that works as a "ruler". By artificially making this ruler molecule shorter or longer, we can make the attachment organelle, a supramolecular structure about 300 nm long, shorter or longer. Kohki, a student in my group when I was at Gakushuin University, and Tsuyoshi at the National Institute of Infectious Diseases made many mutants and did lots of experiments. Many thanks. Kohki received Excellent Presentation Awards at the Annual Meeting of the Japanese Society for Bacteriology and at the meeting of the Japanese Society of Mycoplasmology. The press release for this paper is here.

We found that the bacterium Flavobacterium forms huge swirling patterns when cells gather in groups. For some reason, the swirls always rotate counterclockwise. Since this bacterium lives in soil, if you could peek into the ground, it might be spinning round and round there without us ever knowing. Shoko, a student in my group when I was at Gakushuin University, did most of the hard work on the experiments. See here for movies. The press release is here.

We revealed the unique swimming style of the pathogenic bacterium Campylobacter. Of its two flagella, it wraps one around its body and moves forward like a drill. This was a collaboration with Morgan and Eli. The mutants Eli made were spot on, with the flagella beautifully labeled. Eli stayed in Japan for a while, and we talked about research almost every day over beers. Good memories. That was before COVID, so it feels like a long time ago. The press release is here.

Spiroplasma, a helical bacterium, can move freely even though it has no flagella. Among bacteria, only Spiroplasma and Synechococcus are known to swim without flagella. Spiroplasma twists its body to form two helices and generates propulsion very efficiently, and we showed that the propagation of this twist is passive. Tatsuro, a student in my group when I was at Gakushuin University, worked hard on the experiments. The work was also chosen for the cover of the journal.

We measured the torque of the flagellar motor of Halobacterium, an archaeon. Archaea and bacteria both have a motility machine called the "flagellum", but the two are made of completely different molecules and have different evolutionary origins. There is still a lot we don't know about archaeal flagella. Seiji, a student in my group when I was at Gakushuin, worked hard on the experiments for this study. Come to think of it, Iwata loved horse racing. The Fuchu racecourse is close to UEC. The press release is here.

We revealed how cyanobacteria use "threads" to move like Spider-Man. Thin filaments called type IV pili can extend and retract, driven by a motor at their base. What's more, the cells control this extension and retraction cleverly to move toward light, or conversely to move away from strong light. We also discuss how this works. Nils, Annegret and colleagues reported how cyanobacteria sense the direction of light just ahead of us, but their paper is also very interesting. The cyanobacterial mutants were kindly provided by Masuda at Tokyo Tech. Thank you. Only Nishizaka and I are authors on this paper. Four years after moving to the Nishizaka Lab at Gakushuin University, I finally had a solid result. I remember being really happy. The press release is here.

We studied in detail which proteins make up the motility machinery of the bacterium that causes human pneumonia, and how they are arranged at the nanoscale. Mycoplasma cannot swim. Instead, it moves while stuck to the surface of host cells or glass. This is called gliding motility, and it is thought to be important for Mycoplasma's ability to cause disease. We analyzed dozens of mutants, all of which were made by Kenri at the National Institute of Infectious Diseases. Come to think of it, Musashimurayama, where the institute is located, is also close by. Kenri, please come and visit us.

We showed that Mycoplasma, the smallest living organism, moves by repeating cycles of moving a fixed distance and then stopping. That distance is only about 70 nm (nanometers). That's quite a big step for the smallest organism. It probably moves using something like "legs". For details, please see the website of Prof. Miyata at Osaka City University, the lab where I did my graduate work. I started this research as a graduate student, and Kinoshita, a student in my group when I was at Gakushuin University, developed it much further using an experimental system called the "ghost". He also received the JSPS Ikushi Prize.

We revealed how the bacterium Flavobacterium moves. Remarkably, it seems to crawl over surfaces by making its own surface proteins flow rapidly around and around on the membrane surface. It reminds me of the tracks on a tank. This was work from my postdoc at Nagasaki University. My boss there, Prof. Nakayama, and everyone in the lab took such good care of me. Resources such as mutants and antibodies came from Mark. Thank you. While I stayed in his lab for about two months, I got the result that led to this paper. The experiment finally worked late one night, and I will never forget that moment.

We showed that Mycoplasma has a cytoskeleton shaped like a "jellyfish". Not only its shape but also its component proteins are unique: they are made of proteins closely resembling F1 of ATP synthase. This paper is from my time as a graduate student in the Miyata Lab at Osaka City University. I remember giving an oral presentation abroad with my heart pounding. I was very lucky to get such an interesting result as a graduate student. I think experiences like this are why I became a researcher. Now that I have my own lab, I want to build a great research environment so that new students joining the lab can have experiences like this too.

We showed that bacteria can squeeze through narrow spaces by wrapping their flagella around their cell bodies. Prof. Kan's team built the devices, Wada's team ran the physics simulations, and Kikuchi's team worked on the genetics of the insects and their symbiotic bacteria. This is a result from the whole team of the KAKENHI Transformative Research Areas (B) project "Why Microbes Move". It was made possible by Aoba, a member of the Nakane Lab's first cohort, who did a huge number of experiments. Students also received several presentation awards: 1 2 3 4. We published a UEC news release, posted it on EurekAlert!, and also wrote about it in a blog post.
